How Leucine Triggers Muscle Building

Leucine activates a cellular signaling pathway involving a protein complex called mTOR (mechanistic target of rapamycin), which serves as a key regulator of muscle protein synthesis. Should You Take a Standalone Leucine Supplement?

For most people already consuming adequate complete protein (from whole foods or a quality protein powder) with reasonable leucine content, a separate standalone leucine supplement is unlikely to provide meaningful additional benefit. It may be more relevant for people with very specific circumstances, such as older adults dealing with reduced muscle protein synthesis efficiency (anabolic resistance), where some research has explored whether additional leucine might help offset this age-related change — a conversation best had with a healthcare provider.
